I am planning to travel from Kusadasi to Samos on a Saturday. Would it be important for me to book this online or can I just buy the ticket when I get there ? It is in September so its not the busy season.

I really doubt if you can book online at all.
Better check in slow season there is actually a boat-this is not a regular all year dependable route-they only go when enough people turn up-in the morning on the GR side-to bring them back after a day-excursion.
Buses Izmir (Nearest railhead) _Kus run ev 30 mins or so all day and take about 2 hrs. from the big OTOGAR in Izmir-quite far from the train stations.
