I am new to interrail and the whole concept, but am planning to take a trip across Europe starting from Greece. I wanted to ask for advice what would be the best trip and experience to start from Greece and end Barcelona using interrail, in around 20 days.

This is not a good time for that itinery. Greece is in crisis, after a recent train disaster the network seems to be closed with limited buses running. Even before this it was impossible to travel by train from Greece to another country, the broken links were provided by private bus operators with no acceptance of interrail. The normal way out of Greece is ferry ( Interrail can be used on some, see the Interrail website) to Italy where - but getting to the port is now a problem.

In Italy Interrail is not valid on regional trains which have been contracted out to private operators, nor is it valid on Italo fast trains. It is valid on Trenitalia fast trains but needs reservation, and on Trenitalia/Trenord regional trains without reservation.

Spain can be a problem if you want to travel long distances as the fast trains all require reservations which are basically impossible to obtain outside of Spain. For travel by the high speed train from France to Barcelona you could try getting it through the French system or Interrail App. For travel by local train from the border to Barcelona you don't need reservation.

But "best trip" is not a question that can be answered without a very personal list of requirements.
