Would it be possible to store a large suitcase in a locker for a week while we are on a barge trip?
If so, any idea of the cost per day?

Hi.
Yes it is possible at the station. You can prepay up to 72 hours. After that you have to pay the rest when you return to pick up your luggage: https://rail.cc/passau/passau-central-railway-station/l605
Other option for 2 EUR per day is at the travel office of one of the shipping companies. There you pay 2 EUR per day which should be much cheaper: https://www.donau-schiffs-reise-center.de/index.php/gepaeckaufbewahrung
I also recommend to ask at the company you travel with. Usually they offer such a service to store your luggage at their office.
