Can I catch the train departing at 21:43 and arrive in lisbon at 07:20, but departing on the day that my pass ends?

No.
If your pass ends on the day of departure from Madrid it is only valid until midnight of that day and you would need a second, separate ticket for the remainding part of the route to Lisbon. You would need a reservation for your Interrail ticket plus a regular ticket from the last stop before midnight to Lisbon - it is quite likely that it will be cheaper to buy an advance fare ticket all the way from Madrid to Lisbon [u]https://rail.cc/en/night-train/madrid-lisbon-th-332/71[/u] instead (or travel one day earlier if you want to use your Interrail ticket).
