I live in the UK, I want to get and InterRail Pass for one week on the listed countries (Italy-Austria-Czech Republic), which is the best option?
Also, my gf, who lives in Ecuador, South America, will be coming with me. Can I buy an InterRail Pass for her as well?

The shortest available Interrail pass is the 5in15: [u]https://rail.cc/en/interrail-global-pass[/u] so if you're travelling just a week, this might be not your best option.
Also, your girlfriend would need to buy a Eurail pass, unless her current residency is in Europe: [u]https://rail.cc/en/eurail[/u]
Do you know which cities you want to visit?
I guess it would be cheaper for you to buy single tickets.

OK, in that case, just go with single tickets.
Milano - Verona: Hourly local trains, travel time 01:55, 12,70€, buy locally. Alternative faster Frecciabianca Intercity trains running at least hourly, travel time 01:23, 22€ when bought locally (cheaper advance fares available online).
Verona - Venice: Similarly to Milano - Verona you can choose between slower but cheaper regional trains (01:27, 8,70€) or faster but more expensive Frecciabianca trains (01:10, 23,50€).
Venice - Vienna: Buy online in advance to get cheap fares. Possible by overnight train: [u]https://rail.cc/en/night-train/venice-vienna-en-236/97[/u]
Vienna - Prague: Two hourly direct Railjet trains. Buy online in advance to get cheap fares.
